Histone mutations are associated with some types of cancer, such as gliomas and osteosarcomas. These mutations generally exist in the N-terminal tail or globular domains of histone proteins and some act in a dominant fashion. Certain mutations occur at or nearby post-translationally-modified sites (PTMs), and thus could disrupt the reading, writing and erasing of certain epigenetic marks.
